Derivation

The specimen was taken from a patient prior to chemotherapy for metastatic melanoma.

The COLO 829BL cell line was established at the ATCC by Epstein-Barr virus (EBV) transformation of of a sample of peripheral blood provided by G.E. Moore.

A cell line was also produced from the melanoma, and is available as COLO 829 (ATCC CRL-1974).

Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 to 4 x 105 viable cells/mL. Maintain cell density between 2 x 105 and 2 x 106 viable cells/mL.
Medium Renewal: Every 2 to 3 days
Cryopreservation Complete growth medium, 95%; DMSO, 5%. Cell culture tested DMSO is available as ATCC? Catalog No. 4-X.
